What Is the HPV DNA Test?

The HPV DNA test is a molecular diagnostic test that identifies the genetic material (DNA) of the Human Papillomavirus (HPV) in cervical or genital samples.
It detects both low-risk types that cause warts and high-risk types (especially HPV 16 and 18) that can lead to cervical, vaginal, vulvar, or anal cancers if untreated.

Why Is It Important?

HPV infection is extremely common — up to 80% of sexually active individuals are exposed during their lifetime.
In most cases, the immune system clears the virus naturally; however, persistent infection with high-risk HPV types can cause precancerous lesions (CIN 1–3) and cervical cancer.

The HPV DNA test enables:

  • Early identification of high-risk infections before symptoms appear
  • Targeted follow-up for abnormal Pap smear results
  • Prevention of cervical cancer through timely monitoring and treatment
  • Safe testing for both women and men

When Should It Be Done?

  • As part of routine screening starting from age 30, combined with the Pap smear (co-testing)
  • In women with abnormal cytology (ASC-US, LSIL, HSIL) to clarify risk
  • After treatment for cervical lesions, to confirm viral clearance
  • In cases of visible genital warts, to determine viral type and recurrence risk
  • For men, when genital warts or HPV-related lesions are suspected

Screening interval:

  • Every 5 years with Pap co-testing, or
  • Every 3 years if performed alone (depending on risk profile)

How Is the Test Performed?

  1. A small sample of cells is collected from the cervix (for women) or from the genital/urethral area (for men).
  2. The sample is placed in a liquid medium and analyzed using PCR (Polymerase Chain Reaction) technology.
  3. The test detects specific HPV genotypes, differentiating low-risk and high-risk types.
  4. Results are typically available within 13–15 days.

The procedure is quick, painless, and does not require anesthesia.

Understanding the Results

Negative result: No high-risk HPV types detected — low cancer risk.
Positive result: High-risk HPV types identified — requires follow-up (e.g., Pap smear, colposcopy, or biopsy).

The test report may list individual genotypes (e.g., HPV 16, HPV 18, HPV 31, HPV 33), indicating which types are present and their associated risk level.

Note: A positive result does not mean cancer is present, but it shows that careful monitoring is necessary.

HPV DNA Test vs. Pap Smear

  • Pap smear: Examines the cells of the cervix for changes.
  • HPV DNA test: Detects the virus responsible for those changes.
  • Combined testing (co-testing): Offers the most reliable method for preventing cervical cancer.

While Pap smears can miss some early infections, HPV DNA testing identifies viral presence years before visible cell abnormalities develop.
